目的 :提高难溶性药物丹参酮的体外溶出度。方法 :以预胶化淀粉为载体 ,采用溶剂沉积法将丹参酮制备成了溶剂沉积物。测定了丹参酮、溶剂沉积物以及机械混合物的体外溶出度 ,并通过扫描电镜观测以及红外光谱分析对它们进行了研究。结果 :溶剂沉积物明显提高了丹参酮的溶出度 ;丹参酮以结晶形式沉积于载体表面 ,结晶颗粒与沉积前相比明显减小 ,且出现大量针状微晶 ;丹参酮与载体间仅仅是物理作用。结论 :采用溶剂沉积法提高丹参酮的体外溶出度是可行的。
Objective: To improve the in vitro dissolution rate of the insoluble drug tanshinone. Method :Preparation of tanshinone as a solvent deposit by solvent deposition with pregelatinized starch as carrier. The in vitro dissolution of tanshinones, solvent deposits, and mechanical mixtures was determined, and they were investigated by scanning electron microscopy and infrared spectroscopy. RESULTS: Solvent deposits significantly increased the dissolution of tanshinone. Tanshinone was deposited on the surface of the carrier in the form of crystals. Crystal particles were significantly reduced compared to those before deposition. A large number of acicular crystallites appeared; tanshinone and the carrier only had physical effects. Conclusion: It is feasible to increase the in vitro dissolution of tanshinone by solvent deposition.
