目的:研究依托泊苷(Etoposide,VP-16)磷脂复合物的制备方法并对其理化性质进行分析。方法:采用单因素考察并优化复合物制备工艺,对新生成的物质进行紫外光谱、红外光谱、差示扫描量热分析并研究其溶解性能和油水分配系数。结果:VP-16磷脂复合物的优化制备条件为:四氢呋喃作溶剂,药物与大豆磷脂(Soy Phospholipids,SP)的物质的量比为1∶1.2,25℃搅拌0.5 h;新生成物质的光谱分析不同于反应物单体及VP-16和SP的物理混合物;3种不同pH介质中,VP-16的溶解度和油水表观分配系数均有明显提高。结论:VP-16磷脂复合物可明显提高VP-16的溶解度和油水表观分配系数。
Objective: To study the preparation of phospholipid complex of Etoposide (VP-16) and its physicochemical properties. Methods: The single factor was used to study and optimize the preparation process of complex. The newly formed material was analyzed by UV, FTIR and differential scanning calorimetry and its solubility and oil-water partition coefficient were studied. Results: The optimized preparation conditions of VP-16 phospholipid complex were as follows: THF as solvent, the ratio of drug to Soy Phospholipids (SP) was 1: 1.2, stirring at 0.5 ℃ for 0.5 h; Different from the reactant monomer and the physical mixture of VP-16 and SP, the solubility of VP-16 and the apparent partition coefficient of oil-water in three different pH media were significantly increased. Conclusion: VP-16 phospholipid complex can significantly improve the solubility of VP-16 and the apparent partition coefficients of oil and water.
