目的:通过对自由活动大鼠睡眠时相影响的研究,为百麦安神冲剂改善睡眠,治疗神经衰弱提供实验依据。方法:将埋好电极的大鼠24只随机分成百麦安神冲剂低、中、高剂量组。术后第8d给予药量同等体积的0.5%的羧甲基纤维素钠(CMC-NA)溶液,记录给药前大鼠的脑电(EEG)信号。描记时间为9:00~17:00,共计8 h。次日上午8:30经ig给予实验动物0.675,1.35,2.7 g.kg-1的百麦安神冲剂,连续给药7 d,于末次给药30 min后,对大鼠进行8 h的脑电描记。计算大鼠觉醒(Wake),慢波睡眠1期(SWS1),慢波睡眠2期(SWS2),快动眼睡眠(REMS),总睡眠时间(TST)内的总时间,应用自身配对t检验法进行统计比较。结果:百麦安神冲剂中、低剂量均能显著性延长自由活动大鼠的SWS2期,REMS期和TST;高剂量虽不能显著性延长TST,但其仍显著性延长SWS2期和REMS期。结论:百麦安神冲剂对自由活动大鼠具有改善睡眠的作用。
OBJECTIVE: To provide an experimental basis for improving the sleep and treating neurasthenia by studying the effects of sleeping time in the free-moving rats. Methods: Twenty-four rats with buried electrodes were randomly divided into low, medium and high doses of Baiji Anshen granules. On the 8th day after operation, a solution of 0.5% sodium carboxymethyl cellulose (CMC-NA) in the same volume was given and the electroencephalogram (EEG) signals of the pre-administration rats were recorded. Tracing time is 9: 00 ~ 17: 00, a total of 8 h. The next day at 8:30 am ig to the experimental animals 0.675,1.35,2.7 g.kg-1 of Bai Anshen granules, continuous administration of 7 d, after the last administration of 30 min, the rats were 8 h EEG Tracing. The total time of rats in Wake, SWS1, SWS2, REMS and TST was calculated and their paired t-test Law for statistical comparison. Results: Baishen Anshen granules could significantly prolong the SWS2, REMS and TST of free-moving rats at low and medium doses, while prolonged SWS2 and REMS at high doses did not significantly extend TST. Conclusion: Baiji Anshen granules can improve sleep in free-moving rats.
