目的研究葫芦素B聚乳酸-羟基乙酸毫微球小鼠体内分布。方法小鼠尾静脉给药,HPLC法测定葫芦素B在小鼠血浆、心、肝、脾、肺、肾、脑中的浓度。结果葫芦素B聚乳酸-羟基乙酸毫微球主要分布于肝,给药15 min,肝中聚集达最大,为给药剂量的47.15%。结论葫芦素B聚乳酸-羟基乙酸毫微球可实现肝脏被动靶向,所建立的HPLC法用于葫芦素B的体内分布研究,结果良好。
Objective To study the in vivo distribution of cucurbitacin B polylactic acid-glycolic acid nanospheres. Methods The mice were intravenously administered by tail vein and the concentration of cucurbitacin B in plasma, heart, liver, spleen, lung, kidney and brain was measured by HPLC. Results Cucurbitacin B polylactic acid - hydroxyacetic acid nanospheres were mainly distributed in the liver. After administrated for 15 min, the aggregates reached the maximum in the liver, accounting for 47.15% of the administered dose. Conclusion The cucurbitacin B polylactic acid-glycolic acid nanospheres can achieve passive liver targeting. The established HPLC method was used to study the in vivo distribution of cucurbitacin B with good results.
