目的采用激光显微共聚焦拉曼光谱仪研究卡马西平片晶型并建立定性判别模型。方法建立卡马西平4种晶型的拉曼对照图谱,用OPUS软件聚类分析法判别出国内不同厂家卡马西平片的晶型,再通过建立定性鉴别模型鉴别未知样品。结果聚类分析结果表明,4批卡马西平片均属于晶型Ⅲ,采用一阶导数法和矢量归一化法处理图谱,用因子分析法计算,选择200~500、650~850、1 000~1 100、1 500~1 700 cm~(-1)谱段建立定性模型,经验证,该模型的灵敏度、专属性、阴性预报性和检出效率均良好。结论激光显微共聚焦拉曼光谱仪可用于卡马西平片的晶型判别及实现快速、准确的定性鉴别。
Objective To study the crystal form of carbamazepine by laser scanning confocal Raman spectroscopy and to establish a qualitative discriminant model. Methods Raman maps of carbamazepine four crystal forms were established. The crystal forms of carbamazepine tablets from different manufacturers were identified by OPUS software clustering analysis, and the unknown samples were identified by qualitative identification model. Results The results of cluster analysis showed that four batches of carbamazepine tablets belonged to crystal form Ⅲ. The first derivative and vector normalization were used to process the patterns. The factor analysis method was used to calculate the number of carbamazepine tablets. The results showed that 200 ~ 500, 650 ~ 850, 1000 ~ 1 100,1 500 ~ 1 700 cm ~ (-1) spectrum. The results show that the model has good sensitivity, specificity, negative predictability and detection efficiency. Conclusion Laser microscopy confocal Raman spectroscopy can be used to determine the crystal form of carbamazepine tablets and to achieve rapid and accurate qualitative identification.
