目的:了解糖尿病性白内障晶状体混浊的特点及在此过程中晶状体纤维细胞mdr1a mRNA与P-gp的表达量的变化。方法:将3-4周SD雄性大鼠随机分为A、B两组:A组为对照组,B组腹腔注射链脲佐菌素,观察7,15,21,29 d时大鼠的血糖浓度及体重的变化和晶状体混浊情况、晶状体纤维细胞发生的形态学改变以及P-gp及mdr1a mRNA的表达量的变化。结果:B组大鼠腹腔注射链脲佐菌素7 d后血糖浓度大于16.7 mmol/L,随时间的延长,血糖浓度不断升高,晶状体混浊加重。纤维细胞首先表现为肿胀、细胞内有空泡形成,最终表现为纤维细胞的崩解、破裂。晶状体混浊部位在赤道部距周边囊膜100μm处;大鼠腹腔注射链脲佐菌素7 d后,mdr1a mRNA与P-gp的表达增加,15 d时达到高峰,后逐渐下降。结论:糖尿病性白内障的大鼠晶状体特定部位发生混浊,并随血糖升高混浊逐渐加重,晶状体纤维细胞发生特征性改变;P-gp与糖尿病性白内障发生有关。
OBJECTIVE: To investigate the characteristics of lens opacification in diabetic cataract and the changes of mdr1a mRNA and P-gp expression in lens fibroblasts during this process. Methods: Three to four weeks SD male rats were randomly divided into A, B groups: A group as the control group, B group intraperitoneal injection of streptozotocin, 7,15,21,29 d when the blood glucose Concentration and weight changes and lens opacification, morphological changes of lens fiber cells and P-gp and mdr1a mRNA expression changes. Results: In group B, streptozotocin was intraperitoneally injected for more than 16.7 mmol / L streptozotocin. With the prolongation of time, the blood glucose level increased and the opacity of the lens increased. Fibroblasts first showed swelling, the formation of intracellular vacuoles, the final performance of the collapse of fibrocytes, rupture. The turbidity of the lens was located 100 μm away from the surrounding capsule in the equatorial part. After 7 days of intraperitoneal injection of streptozotocin, the expression of mdr1a mRNA and P-gp increased and reached a peak at 15 days, then decreased gradually. CONCLUSION: The ophthalmopathy of diabetic cataract rats is turbid at certain sites, and with the increase of blood sugar, the opacity of ophthalmocytes gradually aggravates and the changes of lens fiber cells occur. P-gp is associated with the development of diabetic cataract.
