目的探讨组蛋白乙酰化酶p300抑制剂姜黄素对小鼠胚胎心脏早期发育转录因子Tbx5表达的影响。方法取清洁级健康成年昆明小鼠,雌雄合笼,受孕成功后于妊娠期第7.5天(E7.5)开始腹腔注射姜黄素75 mg.kg-1.d-1,分别在胚胎心脏发育不同时间点(E11.5、E14.5和E17.5)断颈处死孕鼠,取胎鼠心脏,提取RNA,实时荧光定量(RT)-PCR检测Tbx5的表达。同时设立二甲基亚砜(DMSO)组(腹腔注射相同剂量DMSO)和空白对照组(不做任何处理)作为对照。结果 1.E11.5时,姜黄素组、DMSO组和空白对照组Tbx5的表达水平分别为2.06±0.16、1.08±0.13和1.08±0.18,姜黄素组Tbx5表达较空白组升高1.90倍,差异有统计学意义(P<0.05)。2.E14.5时,姜黄素组、DMSO组和空白组Tbx5的表达水平分别为1.21±0.08、0.71±0.04和0.64±0.10,姜黄素组Tbx5表达较空白组升高1.88倍,差异有统计学意义(P<0.05)。3.E17.5时,姜黄素组、DMSO组和空白对照组Tbx5的表达分别为1.09±0.07、0.79±0.06和0.75±0.06,姜黄素组Tbx5表达较空白组升高1.45倍,差异有统计学意义(P<0.05)。结论姜黄素能透过胎盘,升高小鼠胚胎心脏早期发育转录因子Tbx5的表达。
Objective To investigate the effect of curcumin, a histone acetylase inhibitor p300, on the expression of transcription factor Tbx5 in early embryonic heart of mouse embryos. METHODS: Healthy Kunming mice of healthy adult age were recruited and the females and males were caged. After the pregnancy was successful, curcumin 75 mg.kg-1.d-1 was injected intraperitoneally on the 7.5th day of pregnancy (E7.5), which was different in embryonic heart development. At the time points (E11.5, E14.5, and E17.5), pregnant rats were killed by cervical dislocation, fetal heart was taken and RNA was extracted. The expression of Tbx5 was detected by real-time fluorescence quantitative RT-PCR. At the same time, a dimethyl sulfoxide (DMSO) group (intraperitoneal injection of the same dose of DMSO) and a blank control group (without any treatment) were set as controls. Results 1. The expression levels of Tbx5 in curcumin group, DMSO group and blank control group were 2.06±0.16, 1.08±0.13 and 1.08±0.18 at E11.5, respectively. The expression of Tbx5 in curcumin group was 1.90 times higher than that in blank group. Statistically significant (P<0.05). 2. At E14.5, the expression levels of Tbx5 in curcumin group, DMSO group and blank group were 1.21±0.08, 0.71±0.04 and 0.64±0.10, respectively. In curcumin group, the expression of Tbx5 was increased 1.88 times compared with blank group. Significance (P<0.05). 3. At E17.5, the expression of Tbx5 in curcumin group, DMSO group and blank control group were 1.09±0.07, 0.79±0.06 and 0.75±0.06, respectively. The expression of Tbx5 in curcumin group was 1.45 times higher than that in blank group. The differences were statistically significant. Significance (P<0.05). Conclusion Curcumin can penetrate the placenta and increase the expression of transcription factor Tbx5 in early embryonic heart of mouse embryos.
