复方中药“天芪航力片”提取物对+Gz致大鼠脑损伤的防护作用

目的:观察复方中药“天芪航力片”提取物对+10 Gz暴露后大鼠脑损伤的影响,进一步探讨其可能的防护作用机制。方法:36只雄性SD大鼠随机分为对照组、+10 Gz组和“天芪航力片”防护组,每组12只。于灌胃给药14 d后,给予+10 Gz、5 min的加速度暴露,观察各组大鼠脑超微结构、脑ATP酶含量、血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量及血液流变学的变化。结果:脑组织电镜和组织化学染色结果显示,与对照组比,+10 Gz组大鼠脑皮层神经细胞出现线粒体肿胀、嵴模糊不清及溶酶体增多等损伤变化,大鼠脑ATP酶含量显著降低(P<0.05);“天芪航力片”防护组脑组织损伤较轻,大鼠脑ATP酶含量无明显变化。与对照组和+10 Gz组比,“天芪航力片”防护组SOD均显著升高(P<0.05)。与对照组相比,+10 Gz组的高、中、低切全血黏度,血浆黏度均显著升高(P<0.05),“天芪航力片”防护组,高、中、低切全血黏度、红细胞压积均显著下降(P<0.05);与+10 Gz组相比,“天芪航力片”防护组,高、中、低切全血黏度、红细胞压积均显著下降(P<0.05)。结论:复方中药“天芪航力片”提取物对+10 Gz暴露引起的脑损伤具有保护作用,其防护机制可能与改善高G暴露引起的大鼠血液流变学改变、降低脂质过氧化损伤及减轻能量代谢障碍有关。 Objective: To observe the effect of compound Chinese medicine “Tianqi Hangli” extract on brain injury induced by +10 Gz in rats and to explore its possible protective mechanism. Methods: Thirty-six male Sprague-Dawley rats were randomly divided into control group, +10 Gz group and “Tianqi Air Force” protective group, 12 rats in each group. After intragastric administration for 14 days, the rats were exposed to +10 Gz for 5 min. The ultrastructure of brain, ATP content, serum SOD activity and malondialdehyde MDA content and hemorheology changes. Results: The results of electron microscopy and histochemical staining showed that compared with the control group, the cortical neurons in the + 10 Gz group showed mitochondrial swelling, blurred cristae and increased lysosome. The content of ATPase (P <0.05). The injury of brain tissue in Tianqi Hangli Tablet ’s protective group was lighter, and there was no obvious change of ATPase in rat brain. Compared with control group and +10 Gz group, the SOD of “Tianqi Air Force” protective group increased significantly (P <0.05). Compared with the control group, whole blood viscosity and plasma viscosity of +10 Gz group were significantly increased (P <0.05), and “Tianqi Air Force” protective group, high, medium and low The whole blood viscosity and hematocrit were significantly decreased (P <0.05). Compared with the +10 Gz group, the “Tianqi Air Force” protective group, the high, middle and low cut whole blood viscosity, hematocrit Decreased significantly (P <0.05). CONCLUSION: The compound Chinese herbal medicine “Tianqi Hangli Tablet” has a protective effect on brain injury induced by +10 Gz exposure. The protective mechanism may be related to the improvement of hemorheological changes and lipid lowering in rats induced by high G exposure Peroxidative damage and reduce energy metabolism disorders related.
