目的:建立黄芪、苦参药对提取物中黄芪甲苷的HPLC-ELSD测定方法。方法:采用蒸发光散射检测器(ELSD)对黄芪、苦参药对提取物中的黄芪甲苷进行HPLC分析。色谱柱:Phenomsil C18(250×4.6mm,5mm),流动相:乙腈一水(32:68),流速:1.0mL·min-1,柱温:30℃。ELSD参数:漂移管温度为41℃,载气压力为3.3bar(1bar=100kPa)。结果:按选定的色谱条件测定,平均加样回收率达99.5%,RSD为l.9%。结论:本方法简便、准确,可用于黄芪、苦参药对提取物中黄芪甲苷的含量测定。
Objective: To establish an HPLC-ELSD method for the determination of Astragaloside Ⅳ in Radix Astragali and Sophora flavescens. Methods: Astragalus membranaceus and Astragalus membranaceus were determined by HPLC with ELSD. Column: Phenomsil C18 (250 × 4.6mm, 5mm), mobile phase: acetonitrile-water (32:68), flow rate: 1.0mL · min-1, column temperature: 30 ℃. ELSD parameters: Drift tube temperature 41 ° C, carrier gas pressure 3.3 bar (1 bar = 100 kPa). Results: According to the selected chromatographic conditions, the average recovery rate of 99.5%, RSD was l.9%. Conclusion: The method is simple and accurate and can be used for the determination of Astragaloside Ⅳ in the extracts of Astragalus and Sophora.
