目的评价TiO2-xNx薄膜托槽的细胞生物相容性,并探讨掺氮含量与薄膜厚度对其生物相容性的影响。方法制备TiO2-xNx薄膜托槽试样,将L929细胞分成薄膜托槽组和对照组,其中薄膜托槽组分成不同氮含量组(氩气/氮气比分别为30∶1、30∶1.5、30∶2)和不同厚度组(溅射时间分别为30、60、90min)。通过对L929细胞进行细胞黏附实验、MTT实验和乳酸脱氢酶(LDH)释放实验评价TiO2-xNx薄膜托槽的细胞生物相容性。采用SPSS 19.0统计软件,通过单因素方差分析对实验数据进行统计学分析。结果 L929细胞在薄膜托槽组和对照组托槽表面的黏附和伸展状态均良好。不同氮含量组(24、48、72h)和厚度组(24、72h)的细胞相对增殖率与对照组之间差异均有统计学意义(P<0.05),所有组别的相对增殖率均在80%以上。随着氮含量的增加,细胞相对增殖率降低;随着厚度的增加,细胞相对增殖率增加。LDH实验结果显示,随着氮含量的增加LDH活性增加,随着厚度的增加LDH活性降低,所有组别之间差异均无统计学意义(P>0.05)。结论 TiO2-xNx薄膜托槽细胞毒性为0级,掺氮含量和薄膜厚度因素与L929细胞生物相容性有关。
Objective To evaluate the biocompatibility of TiO2-xNx thin film brackets and investigate the effect of nitrogen content and film thickness on its biocompatibility. Methods TiO2-xNx thin film brackets samples were prepared, the L929 cells were divided into a thin film brackets group and a control group, wherein the thin film brackets components into different nitrogen content group (argon / nitrogen ratio were 30: 1, 30: 1.5, : 2) and different thickness groups (sputtering time were 30,60,90 min). Cell biocompatibility of TiO2-xNx thin film brackets was evaluated by cell adhesion assay, MTT assay and lactate dehydrogenase (LDH) release assay. Using SPSS 19.0 statistical software, the experimental data were statistically analyzed by one-way ANOVA. Results The adhesion and extension of L929 cells in the brackets of the brackets and control groups were good. The relative proliferation rate of different nitrogen content groups (24,48,72h) and thickness group (24,72h) were significantly different from the control group (P <0.05), and the relative proliferation rates of all groups were 80% or more. With the increase of nitrogen content, the relative proliferation rate of cells decreased; with the increase of thickness, the relative proliferation rate of cells increased. The results of LDH showed that the LDH activity increased with the increase of nitrogen content, and the LDH activity decreased with the increase of the thickness. There was no significant difference among all groups (P> 0.05). Conclusion The cytotoxicity of TiO2-xNx thin film brackets is Grade 0, and the nitrogen-doped and film thickness factors are related to the biocompatibility of L929 cells.
