目的观察乏氧对中性粒细胞体外穿越牙龈上皮模型的影响。方法体外培养人牙龈上皮细胞并构建牙龈上皮模型,自外周血分离纯化中性粒细胞,使其在趋化剂的诱导下穿越牙龈上皮模型,以血平板计数和中性粒细胞髓过氧化物酶活性测定等方法检测中性粒细胞的穿越效率。结果常氧环境下(21%O2),3h内66.8%~78.1%的中性粒细胞完成穿越牙龈上皮模型,但乏氧环境下(1%O2)穿越效率仅23.1%~27.5%。结论体外条件下乏氧抑制中性粒细胞穿越牙龈上皮模型。
Objective To observe the effect of hypoxia on neutrophil transdermal gingival epithelial model in vitro. Methods Human gingival epithelial cells were cultured in vitro and the gingival epithelial cells were constructed. Neutrophils were isolated and purified from peripheral blood. The cells were cultured in the gingival epithelial cells under the induction of chemotactic agents. The blood plate count and neutrophil myeloperoxide Enzyme activity determination methods such as detection of neutrophil traversal efficiency. Results Under normoxic conditions (21% O2), 66.8% ~ 78.1% of neutrophils within 3 h completed the transdermal gingival epithelial model, but the viability of only 1% O2 was 23.1% -27.5% under hypoxia. Conclusion In vitro hypoxia inhibits neutrophils from crossing the gingival epithelium.
