为了检测黄芪多糖(Astragalus polysaccharide,APS)对猪外周血单核细胞来源树突状细胞(dendritic cells,DC)细胞因子分泌情况的影响,分析不同细胞因子的表达差异,试验采用抗体芯片技术对不同处理组(APS组、LPS组)树突状细胞培养上清液中的8种细胞因子进行检测。结果表明:用APS和LPS处理树突状细胞后均可对其细胞因子的分泌产生影响,且和对照组细胞因子表达具有显著差异;APS组和LPS组细胞因子表达谱相似,均高表达IL-8、IL-12和TNF-α,低表达IL-4、IL-6、IL-10和TGFβ1,不同之处在于LPS组高表达IL-1β,而APS组低表达IL-1β。说明APS和LPS均具有刺激树突状细胞成熟并调控其细胞因子分泌的功能。
In order to investigate the effect of astragalus polysaccharide (APS) on the cytokine secretion of dendritic cells (DC) derived from porcine peripheral blood mononuclear cells (PBMCs), the expression of different cytokines was analyzed. 8 kinds of cytokines in the dendritic cell culture supernatant of the treatment group (APS group, LPS group) were detected. The results showed that the dendritic cells treated with APS and LPS both had an effect on the secretion of cytokines and had significant differences with the control group. The expression of cytokines in APS group and LPS group were similar, IL-4, IL-6, IL-10 and TGFβ1 were down-regulated in IL-8, IL-12 and TNF-α, except that IL-1β was highly expressed in LPS group and IL-1β in APS group. It shows that both APS and LPS have the function of stimulating the maturation of dendritic cells and regulating the secretion of cytokines.
