目的 研究碱性成纤维细胞生长因子 (basic fibroblast growth factor,b FGF)在体外对人晶状体上皮细胞增殖的影响及机制探讨。方法 取人晶状体前囊膜培养 ,加入不同浓度b FGF(1μg· L- 1、10 μg·L- 1、10 0 μg·L- 1 ) ,48h后免疫组化测增殖核细胞抗原阳性面积率。取人晶状体前囊膜免疫组化测 b FGF受体。结果 一定浓度 (1~ 10 0 ug· L- 1 )的 b FGF在体外有促人晶状体上皮细胞增殖的作用 ,且呈剂量相关性。人晶状体上皮细胞 b FGF受体表达阳性。结论 b FGF通过受体介导可促人晶状体上皮细胞增殖 ,在维持人晶状体上皮细胞的正常生理和后发障的发病机制中占有重要的地位。
Objective To investigate the effect of basic fibroblast growth factor (b FGF) on the proliferation of human lens epithelial cells in vitro and its possible mechanism. Methods The anterior lens capsule of human lens was taken and added with different concentrations of bFGF (1μg · L -1, 10μg · L -1, 100μg · L -1). After 48 hours, the positive area ratio of proliferating cell nuclear antigen . The anterior capsule of human lens was taken for immunohistochemical detection of bFGF receptor. Results The bFGF with a certain concentration (1-10.0 μg · L -1) could promote the proliferation of human lens epithelial cells in vitro and in a dose-dependent manner. Human lens epithelial cells b FGF receptor positive expression. Conclusion b FGF can promote the proliferation of human lens epithelial cells through receptor mediation, which plays an important role in the pathogenesis of maintaining the normal physiological and post-impairment of human lens epithelial cells.
