目的:探讨磷酸化组蛋白(γH2AX)的表达水平,预测肝癌细胞对于电离辐射的敏感性。方法:以不同剂量X射线照射肝癌细胞系HepG2.215,流式细胞术检测肝癌细胞中表达γH2AX的细胞分数;免疫细胞化学法检测肝癌细胞中γH2AX灶点数,MTT比色法检测放射线照射后的细胞活性。结果:肿瘤细胞经X射线照射后,γH2AX的表达呈剂量依赖性,随照射剂量的增高而增高;γH2AX的表达水平与细胞死亡率呈正相关(r2=0.984,P<0.05),即γH2AX的表达越高,细胞的死亡率越高。结论:γH2AX表达水平预测肝癌细胞对于放射线治疗的敏感性有较高的可行性和可靠性。
Objective: To investigate the expression of phosphorylated histone (γH2AX) and predict the sensitivity of hepatoma cells to ionizing radiation. Methods: HepG2.215 cells were irradiated with different doses of X-rays and the fraction of cells expressing γH2AX was detected by flow cytometry. The number of γH2AX in hepatocellular carcinoma cells was detected by immunocytochemistry. The number of γH2AX in hepatocellular carcinoma cells was detected by MTT assay Cell activity. Results: The expression of γH2AX was increased in a dose-dependent manner with X-ray irradiation. The expression of γH2AX was positively correlated with cell death (r2 = 0.984, P <0.05), that is, the expression of γH2AX The higher the cell’s mortality rate. Conclusion: It is feasible and reliable to predict the sensitivity of hepatoma cells to radiotherapy for γH2AX expression.
