在根瘤菌的选育工作中,通常要做结瘤试验以鉴定豆科作物的结瘤性能。目前采取的土培法、沙培法、水培法和试管洋菜法等结瘤试验方法都存在着一定的缺点。如土培法、沙培法不能连续观察结瘤的全过程,水培法和试管洋菜法则因通气不良而使结瘤缓慢。为了克服这些缺点,我们采用了“塑料袋培养法”,其优点是:材料简单,操作方便;通气性能好,结瘤速度比一般方法快,如紫云英9天左右就开始结瘤,18天左右就能完成结瘤试验;一目了然,便于观察结瘤过程状况。
