以9个甜高粱材料为对象,设0、70、140和210mmol/L 4个NaCl浓度,在人工气候箱中进行发芽试验,分析了不同NaCl浓度对甜高粱发芽的影响,利用相对盐害率的隶属函数值综合评价了耐盐性;用聚丙烯酰胺凝胶垂直板电泳法,研究了不同NaCl浓度对甜高粱苗中POD同工酶的影响。结果表明:盐胁迫下甜高粱发芽率、发芽指数、发芽势、活力指数、芽长、根长、芽鲜质量、根鲜质量均与对照差异显著(P<0.05)。耐盐性强弱顺序为:泰斯>甜132>BJK156>考利>吉甜3>威利>凯勒>戴尔>吉甜2。与对照相比,随着NaCl浓度增加,9个甜高粱材料POD同工酶表达量均逐渐增加,并且威利、甜132、凯勒、吉甜2、泰斯、吉甜3、考利在3个NaCl浓度下均诱导出了新的酶带P1,戴尔在140和210mmol/L浓度下诱导出了新的酶带P1,BJK156仅在210mmol/L浓度下诱导出了新的酶带P1,吉甜3在70mmol/L浓度下酶带P4消失,在140和210mmol/L浓度下又逐渐出现。甜高粱POD同工酶谱的变化与其耐盐性强弱在一定程度上相关。
Nine sweet sorghums were used as materials, four NaCl concentrations of 0, 70, 140 and 210 mmol / L were set up. Germination tests were conducted in an artificial climate chamber. The effects of different NaCl concentrations on the germination of sweet sorghum were analyzed. The salt tolerance was evaluated synthetically. The effects of different NaCl concentrations on the POD isozymes of sweet sorghum seedlings were studied by using polyacrylamide gel vertical plate electrophoresis. The results showed that the germination rate, germination index, germination potential, vigor index, shoot length, root length, fresh bud quality and root fresh weight of sweet sorghum were significantly different from those of the control under salt stress (P <0.05). Salt tolerance of the order of: Tess> sweet 132> BJK156> Cowley> Kat sweet 3> Willie> Keller> Dell> Kat sweet 2. Compared with the control, the POD isozymes of 9 sweet sorghum materials increased gradually with the increase of NaCl concentration, and the expression of POD isozymes of Willie, Sweet 132, Keller, Ji Tian 2, Taisi, Ji Tian 3, A new enzyme band P1 was induced at all three NaCl concentrations. Dell induced a new enzyme band P1 at concentrations of 140 and 210 mmol / L. BJK156 induced a new band P1 at 210 mmol / L, Ji Tian 3 in the 70mmol / L concentration of enzyme disappeared P4, 140 and 210mmol / L concentration gradually emerged. The changes of POD isozymes in sweet sorghum correlated with the salt tolerance of sweet sorghum to a certain degree.
