目的 :探讨三羟异黄酮 (genistein)对异种混合淋巴细胞增殖和凋亡的作用。方法 :建立异种单向混合淋巴细胞培养模型 ,然后分别经 2 0 μmol/Lgenistein和 5 0mg/mlCsA处理 5d ,应用MTT比色法观察各处理组和对照组淋巴细胞增殖改变 ,流式细胞仪检测细胞凋亡。结果 :与CsA组和对照组相比 ,genistein处理组淋巴细胞增殖明显减弱 (P <0 .0 1) ,CsA组和对照组的淋巴细胞增殖无显著性差异 (P >0 .0 5 ) ,genistein处理组、CsA组和对照组的凋亡率分别为 10 .72 % ,2 .2 2 %和 1.89%。genistein处理组的凋亡率明显高于其他两组 (P <0 .0 5 )。结论 :genistein可以抑制异种混合淋巴细胞增殖 ,并促进其凋亡的发生
Objective: To investigate the effect of genistein on the proliferation and apoptosis of heterogeneous mixed lymphocytes. Methods: A heterogeneous unidirectional mixed lymphocyte culture model was established and then treated with 20 μmol / L of genistein and 50 mg / ml of CsA for 5 days respectively. MTT assay was used to observe the proliferation of lymphocytes in each treatment group and control group. Flow cytometry Apoptosis. Results: Compared with CsA group and control group, the proliferation of lymphocytes in genistein group was significantly decreased (P <0.01), while there was no significant difference in lymphocyte proliferation between CsA group and control group (P> 0.05) The apoptosis rates of genistein group, CsA group and control group were 10.72%, 2.2% and 1.89%, respectively. The apoptosis rate of genistein treated group was significantly higher than that of the other two groups (P <0.05). Conclusion: genistein can inhibit the proliferation of heterogeneous mixed lymphocytes and promote their apoptosis
