为研究DF-1细胞的泛素-蛋白酶体系统(UPS)是否参与新城疫病毒(NDV)在细胞中复制的过程。使用Western-blot检测感染NDV Herts/33毒株的细胞样品以判断细胞内泛素化蛋白水平的变化,并检测NDV感染对细胞26S蛋白酶体水解活性的影响。检测泛素-蛋白酶体系统抑制剂对NDV感染后细胞上清中病毒滴度的影响,以及抑制剂MG-132对NDV在细胞中生长状况的影响。结果显示,虽然感染NDV后DF-1细胞内泛素化蛋白水平并未发生明显变化,但26S蛋白酶体的活性显著升高;使用泛素-蛋白酶体系统抑制剂可以抑制NDV在DF-1细胞中的复制。本研究结果证明DF-1细胞内泛素-蛋白酶体系统参与了NDV在DF-1细胞中的复制过程。
To investigate whether DF-1 cells’ ubiquitin-proteasome system (UPS) is involved in the replication of Newcastle disease virus (NDV) in cells. The cell samples infected with NDV Herts / 33 strain were detected by Western-blot to determine the change of intracellular ubiquitinated protein level and the effect of NDV infection on the 26S proteasome hydrolysis activity. The influence of ubiquitin-proteasome inhibitor on the virus titer in supernatant of cell infected with NDV and the effect of inhibitor MG-132 on the growth of NDV in cell were detected. The results showed that, although the ubiquitinated protein level of DF-1 cells did not change significantly after NDV infection, the activity of 26S proteasome increased significantly. Ubiquitin-proteasome inhibitor could inhibit NDV expression in DF-1 cells In the copy. Our results demonstrate that the ubiquitin-proteasome system in DF-1 cells is involved in the replication of NDV in DF-1 cells.
