目的分析FgBβ1689T/G、I6I/D、345C/T和HinfIA/C多态性位点的单体型人群分布特征和脑梗死疾病的关系及其对血浆Fg浓度和分子活性等功能表达的影响。方法选取开滦集团职工1 021人,均留取清晨空腹静脉血测定血糖、尿酸等生化指标及Fg浓度、FMPV/Amax等反映Fg分子聚合功能的指标,并进行基因型。结果 4个多态性位点的等位基因、基因型及其H1、H4、H5、H7单体型的频率在有和无脑梗死组之间无差异性(P>0.05)。以有无脑梗死为因变量的Logistic回归分析显示,性别、饮酒史、口服阿司匹林、高血压病、DBP、BMI、GLU是脑梗死的影响因素。结论 4个多态性位点及其常见单体型均与脑梗死及血浆Fg浓度和分子功能的表达没有明显的直接联系,但阿司匹林可以与H5单体型相互作用,而降低血浆Fg浓度及其分子活性的功能表达。
Objective To analyze the relationship between the distribution of haplotypes and the infarction diseases in FgBβ1689T / G, I6I / D, 345C / T and HinfIA / C polymorphisms and their effects on the functional expression of plasma Fg concentration and molecular activity. Methods A total of 1 021 workers from Kailuan Group were enrolled in this study. Fasting venous blood samples were collected for determination of biochemical and blood glucose, uric acid, Fg concentration, FMPV / Amax, and other indicators reflecting Fg molecular aggregation. Results The alleles, genotypes and haplotypes of H1, H4, H5 and H7 in the four polymorphic loci showed no difference between the groups with and without cerebral infarction (P> 0.05). Logistic regression analysis with or without cerebral infarction showed that gender, history of drinking, oral aspirin, hypertension, DBP, BMI and GLU were the influencing factors of cerebral infarction. Conclusion The four polymorphic loci and their common haplotypes have no direct relationship with cerebral infarction, plasma Fg concentration and molecular function, but aspirin can interact with H5 haplotype and reduce plasma Fg concentration and The functional expression of its molecular activity.
