目的探讨柴芩承气汤(CQCQD)对急性胰腺炎(AP)小鼠血清胆囊收缩素(CCK)-8及胰腺腺泡细胞内钙离子浓度的影响。方法将24只小鼠随机分成对照组、AP组、CQCQD组及siRNA组(n=6)。后3组采用腹腔内注射8%左旋精氨酸溶液(4 g/kg)制作AP小鼠模型。对照组注射相同剂量的生理盐水。造模成功后,CQCQD组予以管喂0.4 mL/100 g体质量的CQCQD,1次/2 h,共3次;siRNA组予以腹腔注射0.88 mg/kgCCK-siRNA,AP组腹腔注射相同体积的生理盐水。观察干预后6 h各组血清CCK-8及胰腺腺泡细胞钙离子浓度及胰腺病理改变。结果AP组血清CCK-8浓度〔(3764.3±369.2)ng/mL〕、胰腺腺泡细胞钙离子荧光强度(34.8±27.1)及胰腺组织病理评分(6.2±1.1)与对照组〔(1253.5±39.5)ng/mL,5.2±2.3,2.8±0.4〕、CQCQD组〔(1230.5±46.1)ng/mL,9.6±1.6,3.8±0.8〕及siRNA组〔(1702.3±598.3)ng/mL,7.6±2.0,4.1±0.5〕相比较差异有统计学意义(P<0.05)。血清CCK-8与胰腺腺泡细胞内钙离子浓度(r=0.793,P=0.021)及胰腺组织病理评分(r=0.847,P=0.000)呈正相关。结论精氨酸诱导AP小鼠模型与血清CCK-8浓度升高、诱发胰腺腺泡细胞钙超载有关,CQCQD可以降低AP小鼠血清CCK-8水平,缓解胰腺腺泡细胞钙超载,减轻胰腺病理改变。
Objective To investigate the effects of Chai Qin Cheng Qi Decoction (CQCQD) on serum levels of cholecystokinin (CCK) -8 and pancreatic acinar cells in acute pancreatitis (AP) mice. Methods Twenty-four mice were randomly divided into control group, AP group, CQCQD group and siRNA group (n = 6). The latter three groups were injected with 8% L-arginine solution (4 g / kg) to make AP mouse model. The control group was injected with the same dose of saline. The rats in CQCQD group were given 0.4 mL / 100 g body weight of CQCQD once a day for 2 times for 3 times. The siRNA group was intraperitoneally injected with 0.88 mg / kg CCK-siRNA, and the AP group was injected intraperitoneally with the same volume of physiology brine. The levels of serum Ca2 +, pancreatic acinar cell and the pathological changes of pancreas were observed 6 h after intervention. Results The serum CCK-8 concentration in the AP group was (3764.3 ± 369.2) ng / mL, and the pancreatic acinar cell calcium fluorescence intensity was 34.8 ± 27.1 and 6.2 ± 1.1 in the pancreatic tissue and 1253.5 ± 39.5 in the control group (1230.5 ± 46.1) ng / mL, 9.6 ± 1.6, 3.8 ± 0.8] and siRNA group [(1702.3 ± 598.3) ng / mL, 7.6 ± 2.0 , 4.1 ± 0.5〕 compared with the difference was statistically significant (P <0.05). Serum CCK-8 was positively correlated with intracellular calcium concentration in pancreatic acinar cells (r = 0.793, P = 0.021) and pancreatic histopathological score (r = 0.847, P = 0.000). CONCLUSION: The arginine-induced AP mouse model is associated with elevated serum CCK-8 concentration and induces calcium overload in pancreatic acinar cells. CQCQD can reduce serum CCK-8 levels in AP mice, relieve pancreatic acinar cell calcium overload and reduce pancreatic pathology change.
