本文应用~(125)IUdR标记K562靶细胞技术,建立体外测定正常人外周血淋巴细胞NK活性的方法。本法成功的关键是K562靶细胞必需用胰酶和DNA酶共同处理后才能测到~(125)IUdR的释放,而不引起未受损靶细胞明显释放~(125)IUdR。此外,研究了影响~(125)IUdR参入靶细胞和反应条件的一些因素。检测了64例正常健康者(男34例,女30例)外周血NK细胞的活性。男性平均值为52.48±9.95%,女性平均值为38.21±12.00%,两者之间有非常显著的差异(P<0.01)。
In this paper, ~ (125) IUdR labeled K562 target cell technology to establish a method for the determination of normal human peripheral blood lymphocytes NK activity. The key to the success of this method is that K562 target cells must be co-treated with trypsin and DNase to measure ~ (125) IUdR release without appreciable release of ~ (125) IUdR in intact target cells. In addition, some factors affecting the incorporation of ~ (125) IUdR into target cells and reaction conditions were investigated. The activity of peripheral blood NK cells in 64 normal controls (34 males and 30 females) was detected. The average value of males was 52.48 ± 9.95%, the average females was 38.21 ± 12.00%, there was a significant difference between the two (P <0.01).
