目的观察医触景粉剂对小鼠免疫功能与抑瘤作用。方法建立H22肝癌小鼠模型,将接种H22肝癌细胞的小鼠随机分为模型对照组、阳性对照组、医触景高、中、低剂量组。观察对小鼠体重、瘤重及抑瘤率的影响;进行ConA诱导的小鼠淋巴细胞转化实验和小鼠NK细胞活性测定,以及小鼠腹腔巨噬细胞吞噬荧光微球试验与小鼠外周血T淋巴细胞亚群和T、B细胞分析。结果医触景中、高剂量组和阳性对照组瘤重减轻,明显低于模型对照组(P<0.05),3个剂量组的抑瘤率分别为30.30%、32.9%和38.06%,呈剂量-效应关系;阳性对照组淋转A差值和NK细胞活性明显低于模型对照组(P<0.05);高剂量组NK细胞活性明显高于模型对照组(P<0.05)。该受试物阳性对照组小鼠腹腔巨噬细胞吞噬荧光微球的吞噬率明显低于模型对照组(P<0.05),其他各剂量组小鼠吞噬率与模型对照组比较差异均无统计学意义(P>0.05)。该受试物各剂量组小鼠外周血T淋巴细胞亚群CD3、CD4和CD8比值与模型对照组比较差异均无统计学意义(P>0.05)。结论医触景粉剂能抑制H22肝癌小鼠实体瘤的生长,具有增强小鼠免疫功能的作用,对小鼠移植性肿瘤具有抑瘤作用。调节荷瘤小鼠的免疫功能可能是其发挥作用的途径之一。
Objective To observe the immune function and anti-tumor effect of YTT powder in mice. Methods Mouse model of H22 hepatoma was established. Mice inoculated with H22 hepatocarcinoma cells were randomly divided into model control group, positive control group, high, medium and low dose group. The effects of ConA on lymphocyte transformation and NK cell activity in mice were observed, as well as the results of mice peritoneal macrophage phagocytosis fluorescent microspheres assay and mouse peripheral blood T lymphocyte subsets and T, B cell analysis. Results The tumor weight of high dose group and positive control group was significantly lower than that of the model control group (P <0.05). The inhibition rates of the three dose groups were 30.30%, 32.9% and 38.06% (P <0.05). The NK cell activity of the high dose group was significantly higher than that of the model control group (P <0.05). The phagocytosis rate of macrophage phagocytosis fluorescent microspheres in the positive control group was significantly lower than that in the model control group (P <0.05), while the phagocytosis rate in the other dose groups was not statistically different from the model control group Significance (P> 0.05). The ratio of CD3, CD4 and CD8 in peripheral blood T lymphocyte subsets in each dose group of the test substance was not significantly different from that in the model control group (P> 0.05). Conclusion YTJ powder can inhibit the growth of H22 hepatocarcinoma in mice, enhance the immune function of mice, and has anti-tumor effect on transplanted tumors in mice. Regulating the immune function of tumor-bearing mice may be one of the ways to play its role.
