通过观察维拉帕米、肝素、地塞米松在体外对牛晶体上皮细胞(BLEC)增殖和细胞内 Ca~(++)的影响评价了它们抑制后囊混浊的价值。结果维拉帕米呈浓度依赖性抑制 BLEC 增殖[P<0.05(10mg/L)或 P<0.01(≥20mg/L)]和降低细胞内 Ca~(++),并能增强肝素的增殖抑制作用;肝素500mg/L 时可明显抑制增殖(P<0.01)和降低细胞内 Ca~(++),500mg/L 以上抑制作用不随浓度增加而增强;地塞米松对 BLEC 的增殖无明显影响,浓度为250~500mg/L 时细胞内 Ca~(++)浓度增高,再加入维拉帕米80mg/L 可使细胞内 Ca~(++)浓度明显下降。为临床选择药物和剂量提供了理论依据。
By observing the effects of verapamil, heparin and dexamethasone on the proliferation and intracellular Ca ~ (++) in bovine lens epithelial cells (BLECs) in vitro, we evaluated the value of their inhibition of posterior capsule opacification. Results Verapamil could inhibit the proliferation of BLEC in a concentration-dependent manner [P <0.05 (10mg / L) or P <0.01 (≥20mg / L)] and decrease the intracellular Ca ~ (++) (P <0.01) and decrease the intracellular Ca ~ (++) concentration above 500mg / L, while the inhibitory effect did not increase with the increasing concentration of 500mg / L heparin. Dexamethasone had no significant effect on the proliferation of BLECs, The concentration of intracellular Ca ~ (++) increased with the concentration of 250 ~ 500mg / L, and the concentration of verapamil 80mg / L decreased the intracellular Ca ~ (++) concentration significantly. Provide a theoretical basis for the clinical choice of drugs and dosage.
