利用发根农杆菌LBA940 2和R16 0 1诱导药用植物何首乌产生毛状根。由PCR扩增和Southern杂交证实 ,由LBA940 2诱导的毛状根整合了Ri质粒的TL DNA片段 ;冠瘿碱的高压纸电泳证实 ,由两种菌诱导的毛状根整合了Ri质粒的TR DNA片段。毛状根经抗生素和温度除菌后 ,表现出粗壮、绒毛密集、分枝多 ,且在无激素的培养基上能够快速稳定繁殖的特性。经筛选得到何首乌毛状根优良离体培养系PC9e和PC1e。
Hairy roots were produced from the root of Radix Polygoni multiflori by using Agrobacterium rhizogenes LBA940 2 and R16 0 1. PCR amplification and Southern blot confirmed that the hairy root induced by LBA940 2 incorporated the TL DNA fragment of Ri plasmid. High pressure paper electrophoresis of Opuntia demonstrated that the hairy roots induced by the two bacteria integrated the TR of Ri plasmid DNA fragment. Hairy roots after antibiotic and temperature sterilization, showed strong, dense villi, branches and more, and hormone-free medium can quickly and stable reproduction characteristics. After screening, the excellent hairy roots of Polygonum multiflorum PC9e and PC1e were obtained.
