目的考察新型雌激素衍生物——大黄酸-雌酮对小鼠长骨转化生长因子-β1(TGF-β1)及其受体TβRⅡ蛋白表达的影响,初步探讨该化合物对骨的作用机制。方法取16 d胚胎小鼠前肢尺骨在BGJb培养基中培养48 h,培养基中分别含有1×10-6mol/L大黄酸-雌酮、雌酚酮、大黄酸及雌酚酮+大黄酸混合物。采用免疫组织化学方法测定长骨骺板静止区、增殖区和肥大区TGF-β1和TβRⅡ基因表达产物。结果与空白对照组相比,雌酚酮组、大黄酸-雌酮和雌酚酮+大黄酸混合组骺板各区TGF-β1和TβRⅡ蛋白水平明显上调。大黄酸组仅肥大区TβRⅡ上调,其他各区TGF-β1和TβRⅡ蛋白水平均无明显改变。与雌酚酮组相比,大黄酸-雌酮组静止区和增殖区TGF-β1和TβRⅡ的表达均增强。结论培养基中加入大黄酸-雌酮可上调长骨TGF-β1及其受体的表达,并可能与其促进体外培养胚胎小鼠长骨生长有关。
Objective To investigate the effect of a new estrogen derivative, rhein-estrone, on the expression of transforming growth factor-β1 (TGF-β1) and its receptor TβRⅡ in long bone of mice and to explore the mechanism of action of this compound on bone. Methods The ulnar forelimbs of embryonic mice were cultured for 48 h in BGJb medium on day 16, and the media contained 1 × 10-6 mol / L rhein-estrone, estrone, rhein and estrone + rhein mixture . Immunohistochemistry was used to detect the expression of TGF-β1 and TβRⅡgene in resting area, proliferative area and hypertrophied area of long epiphyseal plate. Results Compared with the blank control group, the levels of TGF-β1 and TβRⅡ were significantly up-regulated in the epiphyseal plate of the estrone group, rhein-estrone and estrone + rhein combination groups. In rhein group, only TβRⅡ was upregulated in hypertrophied area, but there was no significant change in TGF-β1 and TβRⅡ protein levels in other areas. Compared with estrone group, rhein-estrone group increased the expression of TGF-β1 and TβRⅡ in quiescent and proliferative areas. Conclusion The addition of rhein-estrone to the medium can up-regulate the expression of TGF-β1 and its receptor, and may be related to the growth of long bones in vitro.
