目的比较前列腺癌(PCa)和前列腺增生(BPH)患者及健康体检者外周血中Th17细胞、Treg细胞的表达差异,初步探讨PCa发生的免疫学机制。方法收集本院泌尿外科2014年1月-2016年12月收治的PCa患者45例(PCa组),42例同期BPH患者为BPH组,另选取同期32例健康体检者为对照组。采用Facs CantoⅡ流式细胞仪检测3组对象外周血Th17细胞和Treg细胞比例,分析PCa患者血清T-PSA与Treg细胞、Th17细胞相关性。结果与对照组及BPH组相比,PCa组外周血中Treg、Th2均明显升高,而Th1细胞、Th17细胞比例均下降,差异均有统计学意义(P<0.05);PCa患者外周血浆T-PSA与调节T细胞比例呈正相关(r=0.78,P<0.05),与Th17细胞呈负相关(r=-0.63,P<0.05)。结论 Th17、Treg细胞可能参与PCa发生发展的病理过程,Th17/Treg细胞检测有助于PCa的早期诊断。
Objective To compare the expression of Th17 cells and Treg cells in peripheral blood of patients with prostate cancer (PCa) and benign prostatic hyperplasia (BPH) and healthy subjects and to investigate the immunological mechanism of PCa. Methods 45 cases of PCa patients (PCa group) admitted from January 2014 to December 2016 in our urology department were enrolled. In the same period, 42 BPH patients were selected as BPH group. Another 32 healthy controls were selected as the control group. Facs Canto Ⅱ flow cytometry was used to detect the ratio of Th17 cells to Treg cells in peripheral blood of three groups. The correlation between T-PSA, Treg cells and Th17 cells in PCa patients was analyzed. Results Compared with the control group and the BPH group, the levels of Treg and Th2 in peripheral blood of PCa group were significantly increased, while the levels of Th1 and Th17 cells in PCa group were significantly decreased (P <0.05). The levels of T -PSA was positively correlated with the proportion of regulatory T cells (r = 0.78, P <0.05), but negatively correlated with Th17 cells (r = -0.63, P <0.05). Conclusion Th17 and Treg cells may be involved in the pathogenesis of PCa. Th17 / Treg cells may be helpful for the early diagnosis of PCa.
