目的观察慢性粒细胞白血病(CML)患者葡萄糖神经酰胺合酶(GCS)和多药耐药1(MDR-1)基因的表达变化及其意义。方法建立实时荧光逆转录聚合酶链反应(RT-PCR)检测GCS、MDR-1和磷酸甘油醛脱氢酶(GAPDH)基因的方法,检测CML初发组25例和急变组25例患者外周血GCS、MDR-1基因表达水平,以GAPDH为内参基因,两组基因表达水平的差异采用△△CT相对定量法。结果外周血GCS水平CML急变组较初发组显著增高(4.84倍,P<0.01),MDR-1表达CML急变组9例阳性(36%),CML初发组均为阴性。结论GCS和MDR-1基因高表达在CML急变患者的多药耐药中起着重要作用,其水平检测可为CML急变预后判断的新指标。
Objective To observe the changes and significance of glucosylceramide synthase (GCS) and multidrug resistance 1 (MDR-1) gene in patients with chronic myeloid leukemia (CML). Methods The real-time fluorescent reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of GCS, MDR-1 and GAPDH gene in peripheral blood of 25 newly diagnosed CML patients and 25 acute hepatitis patients GCS, MDR-1 gene expression, GAPDH as an internal control gene, the two groups of gene expression differences using △△CT relative quantification. Results The levels of GCS in peripheral blood were significantly increased in CML group (4.84-fold, P <0.01). The positive rate of MDR-1 in CML group was 9% (36%). Conclusion The high expression of GCS and MDR-1 gene plays an important role in the multidrug resistance of patients with acute CML. The detection of GCS and MDR-1 may be a new indicator of the prognosis of CML.
