应用聚合酶链式反应加单链多态性分析(PCR-SSCP)及流式细胞木和细胞免疫荧光技术检测52倒尿路上皮肿瘤K-ras基因突变及rasP21蛋白表达和DNA含量变化,研究ras突变和rasP21蛋白表达及DNA倍体异常与尿路上皮肿瘤预后关系。K-ras基因突变5例(9.65),rasP21蛋白阳性表达36例(69.2%);异倍体22例(42.3%),二倍体30例(57.7%);DNA倍体异常和rasP21蛋白表达随肿瘤病理分级和分期的上升而增加,与肿瘤预后呈正相关(P<0.01);异倍体肿瘤其rasP21蛋白表达率明显高于二倍体肿瘤(P<0.01)。DNA倍体异常和rasP21蛋白表达对尿路上皮肿瘤预后有预测意义。
Polymerase chain reaction plus single-strand polymorphism (PCR-SSCP) and flow cytometry and immunofluorescence were used to detect K-ras gene mutation and rasP21 protein expression and DNA content in 52 inverted urothelial tumors Relationship between ras mutation, rasP21 protein expression and DNA ploidy abnormality and urothelial cancer prognosis. RasP21 protein was positive in 36 cases (69.2%); aneuploidy in 22 cases (42.3%), diploid in 30 cases (57.7%); DNA ploidy abnormality and rasP21 protein expression (P <0.01). The expression of rasP21 protein in aneuploid tumors was significantly higher than that in diploid tumors (P <0.01). DNA ploidy abnormalities and rasP21 protein expression of prognosis of urothelial tumors have prognostic significance.
