当前,对于中药的药物代谢动力学研究主要是对一些能取得单体有效成份的中药进行的。一般是采用灵敏可靠的化学方法测定不同时间的血药浓度,经房室模型拟合或统计矩计算,求出药动学参数。但是,多数中药成份不明,难以得到单体或无法以化学方法测定。况且中药成份复杂,临床又多用复方,对某一单一成份的研究结果并不能反映该药(或制剂)的真实体内过程。因此,在中药研究中以药效为指标的药动学研究方法有特殊的意义。 在一定条件下,体内药量与药物的效应有一定的对应关系,从药效的变化可以推知不同时间内体内药量变化。这种方法虽然受生物个体差异影响,误差较大,但却具有简便易行、便于推广的优点,尤其对于无法进行化学测定的中药,可以提供一些有价值的药动学参数。
At present, the pharmacokinetics study of traditional Chinese medicine is mainly carried out on some traditional Chinese medicines that can obtain monomeric effective ingredients. In general, sensitive and reliable chemical methods are used to determine the blood drug concentration at different times. The pharmacokinetic parameters can be obtained through room model fitting or statistical moment calculation. However, most Chinese medicine ingredients are unknown and it is difficult to obtain monomers or chemically determined. In addition, the complexity of traditional Chinese medicines and the use of compound preparations in clinical practice do not reflect the true in vivo process of the drug (or preparation). Therefore, pharmacokinetics research methods with pharmacodynamics as an indicator in Chinese medicine research have special significance. Under certain conditions, there is a corresponding relationship between the drug dose in vivo and the effect of the drug. From the change of drug efficacy, it can be inferred that the drug dose changes in vivo. Although this method is affected by individual differences, the error is large, but it has the advantage of being simple and easy to promote, especially for the traditional Chinese medicine that can not be chemically determined, it can provide some valuable pharmacokinetic parameters.
