目的:研究四逆汤及其不同配伍方对阿霉素诱导大鼠心力衰竭心肌能量代谢的影响。方法:采用腹腔注射阿霉素(2.5 mg/kg)法,每周1次,共6周,建立大鼠慢性心力衰竭模型。将造模成功大鼠随机分为模型组、附子组(1.5 g/kg)、附子-甘草组(3 g/kg)、附子-干姜组(2.5 g/kg)、四逆汤组(4 g/kg)及卡托普利阳性药组(6.25 mg/kg),并设正常对照组。各治疗组连续灌胃给药4周,模型组和正常组予同体积蒸馏水。采用高效液相色谱法测定各组心肌磷酸腺苷(ATP、ADP和AMP)的含量变化。结果:与正常组比较,模型组大鼠ATP含量明显减少(P<0.01),AMP和ADP含量升高(P<0.05);附子-甘草组、附子-干姜组、四逆汤组和卡托普利组较模型组大鼠心肌ATP含量增加(P<0.05),AMP和ADP含量降低(P<0.05)。结论:四逆汤及其不同配伍方可有效调节心衰大鼠的心肌能量代谢紊乱,延缓慢性心衰的发展,以四逆汤全方效应更为明显。
Objective: To study the effects of Sini decoction and its different prescriptions on doxorubicin-induced myocardial energy metabolism in heart failure rats. Methods: Chronic heart failure model was established by intraperitoneal injection of doxorubicin (2.5 mg / kg) once a week for 6 weeks. The model rats were randomly divided into three groups: model group, aconite group (1.5 g / kg), aconite and licorice group (3 g / kg) g / kg) and captopril positive group (6.25 mg / kg), and normal control group. The treatment groups were given gavage for 4 weeks, and the model group and the normal group were given the same volume of distilled water. The contents of ATP, ADP and AMP in each group were determined by HPLC. Results: Compared with the normal group, the content of ATP in the model group decreased significantly (P <0.01) and the content of AMP and ADP increased (P <0.05); the aconite-licorice group, the aconite-dried ginger group, the Sini Decoction group and the card Compared with the model group, the ATP content in the myocardium of Toptop group was increased (P <0.05) and the content of AMP and ADP was decreased (P <0.05). Conclusion: Sini Decoction and its different compatibility prescriptions can effectively regulate myocardial energy metabolism disturbance in heart failure rats and delay the development of chronic heart failure. The effect of Sini Decoction is more obvious.
