观察血管内膜损伤后 c- m yc反义 RNA在抑制血管平滑肌细胞的增生、迁移及防止动脉粥样硬化形成和血管再狭窄中的作用。选雄性日本大白兔 36只 ,经球囊造成腹主动脉内膜损伤的同时 ,局部给予 c- m yc反义 RNA、正义 RNA及盐水 ,高胆固醇喂养 1 2周处死。测量内膜厚度 ,计数增生细胞核抗原 PCNA阳性细胞数。结果显示 :盐水及正义对照组内膜明显增厚 (31 7μm ) ,PCNA核阳性细胞数明显增多 (6 3% ) ,可见明显的粥样斑块灶形成。而反义 RNA治疗组内膜增厚仅为 2 1 7μm,PCNA阳性细胞为 30 .5 % ,明显低于对照组 ,P<0 .0 1。结果表明 :c- m yc反义 RNA具有显著抑制 VSMC增生 ,减轻粥样斑块形成的作用
To observe the role of c-m yc antisense RNA in inhibiting the proliferation and migration of vascular smooth muscle cells and preventing atherosclerosis and vascular restenosis after vascular intimal injury. Thirty-six male Japanese white rabbits were selected, and c-m yc antisense RNA, sense RNA and saline were given locally to the abdominal aorta by balloon injury. The animals were sacrificed 12 weeks after high cholesterol feeding. The thickness of the intima was measured and the number of proliferating cell nuclear antigen PCNA positive cells was counted. The results showed that the intimal thickening (31 7 μm) and the number of PCNA nuclear positive cells in the saline and normal control group were significantly increased (63%), showing obvious atherosclerotic plaque formation. The antisense RNA treatment group, intimal thickening only 21.7μm, PCNA positive cells was 30.5%, significantly lower than the control group, P <0.01. The results showed that: c-m yc antisense RNA significantly inhibited VSMC proliferation, reduce atherosclerotic plaque formation
