目的探讨培美曲塞联合顺铂对人结肠癌细胞SW480增殖和凋亡的影响。方法 SW480细胞单用培美曲塞(A组)、顺铂(B组)或二药联用(AB组)处理,采用MTT法检测细胞增殖抑制率,流式细胞术检测细胞周期及凋亡率,蛋白质印迹法检测增殖细胞核抗原(PCNA)和凋亡相关蛋白B细胞淋巴瘤/白血病2(Bcl-2)表达。结果培美曲塞和顺铂均呈剂量依赖性抑制SW480细胞的增殖,二药联用作用更为明显。AB组(77.8±2.3)%的SW480细胞被阻滞于G1期,高于A组的(64.9±2.2)%和B组的(66.3±1.1)%(P<0.05)。AB组SW480细胞凋亡率为(21.8±2.3)%,高于A组的(8.5±0.5)%和B组的(11.2±2.1)%(P<0.05)。三组SW480细胞中PCNA和Bcl-2蛋白表达量均较处理前下降(P<0.05)。结论培美曲塞和顺铂均可抑制结肠癌细胞SW480的增殖,并促进其凋亡;二药联用作用更明显。
Objective To investigate the effects of pemetrexed combined with cisplatin on the proliferation and apoptosis of human colon cancer cell line SW480. Methods SW480 cells were treated with pemetrexed alone (group A), cisplatin (group B) or combination of two drugs (group AB). The cell proliferation inhibition rate was detected by MTT assay. Cell cycle and apoptosis were detected by flow cytometry The expression of PCNA and Bcl-2 protein were detected by Western blotting. Results Both pemetrexed and cisplatin inhibited the proliferation of SW480 cells in a dose-dependent manner, and the combination of the two drugs was more obvious. SW480 cells in group AB (77.8 ± 2.3)% were arrested in G1 phase, higher than 64.9 ± 2.2% in group A and 66.3 ± 1.1% in group B (P <0.05). The apoptosis rate of SW480 cells in AB group was (21.8 ± 2.3)%, higher than that in group A (8.5 ± 0.5)% and group B (11.2 ± 2.1)% (P <0.05). The expression of PCNA and Bcl-2 protein in three SW480 cells decreased compared with that before treatment (P <0.05). Conclusion Both pemetrexed and cisplatin can inhibit the proliferation of colon cancer cell SW480 and promote its apoptosis; the combination of the two drugs is more obvious.
