目的:探讨micro RNA-125b(mi R-125b)在口腔鳞状细胞癌(OSCC)患者血浆中的表达及临床意义。方法:用茎环法反转录(RT)-实时荧光PCR法检测85例OSCC患者和46例健康人血浆中mi R-125b的表达水平,评估其作为OSCC诊断标志的检测效能。采用SPSS17.0软件包中的单因素方差分析和ROC曲线分析对结果进行统计学处理。结果:反转录PCR法可特异性扩增血浆中的mi R-125b,与正常对照组相比,OSCC患者组血浆mi R-125b表达水平显著上调(P<0.05);ROC曲线分析显示,血浆mi R-125b水平可有效区分OSCC患者和正常人群,其曲线下面积(AUC)达到0.966;当确定最佳截断值为3.46(fold值)时,血浆mi R-125b对OSCC的诊断敏感度和特异度分别达到89.4%和93.5%。结论:鉴于其良好的诊断效能,血浆mi R-125b有望成为具有潜在应用价值的OSCC生物学标志物。
Objective: To investigate the expression of micro RNA-125b (mi R-125b) in the plasma of patients with oral squamous cell carcinoma (OSCC) and its clinical significance. Methods: The plasma levels of mi R-125b in 85 patients with OSCC and 46 healthy controls were detected by stem-loop reverse transcription-real-time PCR. The detection of mi R-125b was used as a diagnostic marker for OSCC. The results were statistically analyzed using one-way ANOVA and ROC curve analysis in the SPSS17.0 software package. Results: Reverse transcriptase polymerase chain reaction (PCR) could specifically amplify mi R-125b in plasma. Compared with normal control group, the expression of mi R-125b in OSCC patients was significantly increased (P <0.05) The plasma level of mi R-125b was effective in differentiating patients with OSCC from normal subjects with an area under the curve (AUC) of 0.966. The diagnostic sensitivity of the plasma mi R-125b to OSCC was determined when the optimal cutoff value was 3.46 (fold value) And specificity of 89.4% and 93.5% respectively. CONCLUSIONS: Given its good diagnostic efficacy, plasma mi R-125b is expected to be a potential OSCC biomarker.
