目的:观察香连丸对化疗相关性腹泻(CID)小鼠生存时间、腹泻程度、肠道病理损伤、回肠组织中促炎细胞因子TNF-α和IFN-γ的影响,探讨其防治CID的可能机理。方法:采用腹腔注射伊立替康法制备CID裸鼠模型;小鼠随机分为正常组、模型组、洛哌丁胺组、香连丸组。观察裸鼠生存时间、腹泻情况、病理改变及回肠组织中TNF-α和IFN-γ表达情况。结果:与模型组比较,香连丸组生存时间延长(P<0.01),腹泻评分及小肠病理损伤程度均较明显降低(P<0.01),回肠组织中TNF-α、IFN-γ的含量均显著降低(P<0.05,P<0.01)。结论:香连丸对CID具有一定的防治作用,其机制可能与其下调回肠组织中促炎细胞因子TNF-α、IFN-γ的表达有关。
Objective: To observe the effect of Xiang Lian Wan on the survival time, diarrhea, intestinal pathological damage, proinflammatory cytokines TNF-α and IFN-γ in mice with chemotherapy-associated diarrhea (CID), and to explore its possible control of CID mechanism. Methods: The model of CID nude mice was established by intraperitoneal injection of irinotecan. The mice were randomly divided into normal group, model group, loperamide group and Xianglian Wan group. The survival time, diarrhea, pathological changes and expression of TNF-α and IFN-γ in ileal tissue were observed. Results: Compared with model group, the survival time of Xianglian pill group was longer (P <0.01), diarrhea score and the pathological damage of small intestine were significantly lower (P <0.01), the content of TNF-α and IFN-γ in ileum Significantly decreased (P <0.05, P <0.01). CONCLUSION: Xiang-Lian Wan has some preventive and therapeutic effects on CID, and its mechanism may be related to down-regulating the expression of pro-inflammatory cytokines TNF-α and IFN-γ in ileum.
