柴胡总皂甙对运动性疲劳大鼠中枢单胺类神经递质及其代谢产物含量的影响

目的:研究柴胡总皂甙对运动性疲劳大鼠海马、下丘脑单胺类神经递质及其代谢产物含量的影响。方法:将36只雄性Sd大鼠,随机均分为空白组、模型对照组和柴胡总皂甙组,共3组,每组12只。柴胡总皂甙组每天灌胃给药50mg/kg,模型对照组和空白对照组灌胃等量生理盐水,柴胡总皂甙组和模型组大鼠在灌胃1h后进行中等运动强度的水平跑台运动,速度为15m/min,坡度0°,跑台20min,间歇40min,再跑台20min,每天1次,连续14d。末次运动后处死大鼠并分离海马及下丘脑,HPLC-ECD法检测其单胺类神经递质及其代谢产物含量。结果:与空白组比较,模型组大鼠海马区5-HT、5-HIAA、NE含量升高,DA、DOPAC、MHPC含量降低;下丘脑区5-HT、5-HIAA含量升高,DA、DOPAC、NE、MHPC含量降低。与模型组比较,柴胡总皂甙组大鼠海马区5-HT、5-HIAA、NE含量降低,DA、DOPAC、MHPC含量升高;下丘脑区5-HT、5-HIAA含量降低,DA、DOPAC、MPHC含量升高。结论:柴胡总皂甙抗运动性疲劳,可改善运动性疲劳中枢单胺类神经递质合成与代谢紊乱的状况,可作为主要成分广泛应用于抗疲劳食品和药品的市场开发中。 Objective: To study the effects of saikosaponins (SAC) on monoamine neurotransmitters and their metabolites in hippocampus and hypothalamus of rats with exercise-induced fatigue. Methods: 36 male Sd rats were randomly divided into blank group, model control group and saikosaponins group, a total of 3 groups of 12 rats. Bupleurum total saponin group administered intragastrically 50mg / kg, the model control group and the blank control group were given the same amount of normal saline, Bupleurum saponin group and model group rats were given 1h after the level of moderate exercise intensity level Taiwan exercise, the speed of 15m / min, gradient 0 °, treadmill 20min, intermittent 40min, and then run 20min, 1 time per day for 14 days. After the last exercise, rats were sacrificed and the hippocampus and hypothalamus were isolated. The levels of monoamine neurotransmitters and their metabolites were determined by HPLC-ECD. Results: Compared with the blank group, the contents of 5-HT, 5-HIAA and NE in the hippocampus and the contents of DA, DOPAC and MHPC in the model group decreased; the levels of 5-HT and 5-HIAA in the hypothalamus increased, DOPAC, NE, MHPC content decreased. Compared with model group, the content of 5-HT, 5-HIAA and NE in hippocampus and the contents of DA, DOPAC and MHPC increased in hypothalamic rats, while the content of 5-HT and 5-HIAA in hypothalamus decreased, DOPAC, MPHC content increased. Conclusion: Bupleurum saponin against exercise-induced fatigue can improve the synthesis and metabolic disorders of monoamine neurotransmitters in exercise-induced fatigue. It can be widely used in the market development of anti-fatigue foods and pharmaceuticals as the main component.
