目的 研究周围神经损伤后 ,局部应用碱性成纤维细胞生长因子 (basic fibroblast growth factor,b FGF)对脊髓前角运动神经元的保护作用。方法 切断 SD大鼠右侧坐骨神经 ,近端套接单盲硅胶管后 ,管内立即注入 2 0μl b FGF(浓度为 40 0 0 u/ ml)或等量生理盐水。术后 2周 ,观察 L 3- L 5脊髓前角外侧群大、中小型神经元乙酰胆碱酯酶 (acetylcholinesterase,ACh E)、酸性磷酸酶 (acid phosphatase,Acp)和一氧化氮合酶 (nitric oxide synthase,NOS)的阳性细胞数、反应强度和平均灰度。结果 坐骨神经损伤后 ,脊髓腰节段运动神经元因受损伤的影响发生明显退变 ,表现为 ACh E活性降低 ,Acp、 NOS活性升高。局部应用 b FGF后 ,大型运动神经元 (α运动神经元 )阳性细胞数、反应强度和平均灰度接近正常侧 ,与对照组比较具有显著性意义 (P<0 .0 1)。结论 b FGF可以有效地防止周围神经损伤引起的脊髓前角α运动神经元退变 ,促进其恢复
Objective To study the protective effect of basic fibroblast growth factor (b FGF) on motor neurons in anterior horn of spinal cord after peripheral nerve injury. Methods The right sciatic nerve of SD rats was severed, and a single blind silicone tube was inserted into the proximal part of the SD rats. Immediately injected with 20 μl of bFGF (concentration of 40 000 u / ml) or normal saline. Two weeks after operation, the expressions of acetylcholinesterase (AChE), acid phosphatase (Acp) and nitric oxide synthase (nitric oxide synthase) in large and small neurons of lateral and anterior spinal cord of L 3- synthase, NOS) positive cells, response intensity and average grayscale. Results After sciatic nerve injury, the motor neuron in the lumbar spinal cord segment of the spinal cord degenerated obviously. The activity of ACh E was decreased and the activity of Acp and NOS was increased. After local administration of bFGF, the numbers of positive cells, reaction intensity and average gray value of large motor neurons (α motor neuron) were close to normal, which were significantly different from those of the control group (P <0.01). Conclusions b FGF can effectively prevent degeneration of α motor neuron in anterior horn of spinal cord induced by peripheral nerve injury and promote its recovery
