目的:研究三七皂苷Fc对2型糖尿病db/db小鼠肾脏的保护作用。方法:8周龄雄性db/db及其同窝对照db/m小鼠分为db/m对照组,db/db模型组及db/db模型+三七皂苷Fc治疗组,每组5只。三七皂苷Fc组给予三七皂苷Fc 5 mg/(kg·d)橄榄油溶液灌胃,db/m对照组,db/db模型组给予等量橄榄油灌胃,连续8周。灌胃8周末测定小鼠体质量、血糖,收集24 h尿液,并留取血液及肾脏组织,检测尿白蛋白浓度、血肌酐及肾脏病理损伤,并采用免疫组织化学方法检测肾组织整合素连接酶(Integrin Linked Kinase,ILK)表达。结果:与db/db模型组比较,三七皂苷Fc组尿白蛋白明显降低(P<0.05),HE染色显示三七皂苷Fc可明显改善肾脏病理损伤。此外,三七皂苷Fc还可明显下调db/db小鼠肾组织ILK表达。结论:三七皂苷Fc可降低2型糖尿病db/db小鼠尿蛋白,延缓肾脏病变进展,具有肾脏保护作用。
Objective: To study the protective effect of notoginsenoside Fc on the kidney of type 2 diabetic db / db mice. Methods: Eight weeks old male db / db and its littermate db / m mice were divided into db / m control group, db / db model group and db / db model + notoginsenoside Fc treatment group, with 5 rats in each group. Notoginsenoside Fc group was given notoginsenoside Fc 5 mg / (kg · d) olive oil solution gavage, db / m control group, db / db model group given the same amount of olive oil for 8 weeks. At 8 weeks after gavage, the body weight and blood glucose of mice were measured. Urine was collected for 24 hours. Blood and kidneys were collected for determination of urinary albumin, serum creatinine and renal pathological changes. Immunohistochemistry was used to detect the levels of integrin Expression of Integrin Linked Kinase (ILK). Results: Compared with the db / db model group, urinary albumin was significantly decreased in the notoginsenoside Fc group (P <0.05). HE staining showed that notoginsenoside Fc significantly improved renal pathological injury. In addition, notoginsenoside Fc can significantly reduce the db / db mouse kidney ILK expression. Conclusion: Notoginsenoside Fc can reduce urinary protein in type 2 diabetic db / db mice, delay the progression of renal disease, and have renal protective effect.
