目的:研究苇茎汤对人肺腺癌A549细胞凋亡的影响。方法:将苇茎汤培养液浓度稀释为0.25、2.5、25g/L,与A549细胞共同培养,AO∕EB荧光染色观察细胞凋亡形态学改变;流式细胞术分析细胞周期和细胞凋亡率;Real-time PCR检测EGFR、STAT3基因的表达。结果:0.25~25g/L苇茎汤能诱导A549细胞发生凋亡形态学改变,且细胞凋亡率随苇茎汤浓度增加而升高;苇茎汤可将A549细胞周期阻滞于G0/G1期;Real-time PCR结果表明,苇茎汤可以下调A549细胞中EGFR、STAT3基因的表达。结论:苇茎汤可能通过调控EGFR、STAT3信号通路诱导人肺腺癌A549细胞发生早期凋亡。
Objective: To study the effect of Weijin Decoction on the apoptosis of human lung adenocarcinoma A549 cells. Methods: The concentration of reed stem broth was diluted to 0.25, 2.5 and 25g / L, and cultured with A549 cells. AO / EB staining was used to observe the morphological changes of apoptosis. Conclusion: Reed Stem Decoction may induce early apoptosis of human lung adenocarcinoma A549 cells through the regulation of EGFR and STAT3 signaling pathways.
