目的:探讨大蒜素对胰腺癌细胞上皮间质转化(EMT)的影响。方法:常规培养胰腺癌细胞株PANC-1细胞,分别以10μg·L~(-1)转化生长因子-β_1(TGF-β_1),10μg·L~(-1)TGF-β_1+5 mg·L~(-1)大蒜素,5 mg·L~(-1)大蒜素处理PANC-1细胞,另设空白组,镜下观察细胞形态学改变。细胞划痕实验检测大蒜素是否能降低胰腺癌细胞的迁移能力;免疫荧光细胞化学法及免疫印迹法(Western blot)检测各组钙黏附蛋白-E(E-cadherin),波纹蛋白(Vimentin)的表达变化。结果:TGF-β_1能够刺激PANC-1细胞发生典型的EMT形态变化,并伴有E-cadherin表达的上调及Vimentin的下调。划痕实验表明大蒜素能降低胰腺癌PANC-1细胞的迁移能力。免疫荧光细胞化学法及Western blot结果显示,与空白组及TGF-β_1组比较,大蒜素组的E-cadherin表达明显上调(P<0.05),Vimentin明显下调(P<0.05),这表明大蒜素具有抑制PANC-1细胞产生EMT的效应。结论:大蒜素可抑制胰腺癌细胞发生EMT,进而可能在抑制肿瘤浸润与转移过程中发挥重要作用。
Objective: To investigate the effect of allicin on epithelial-mesenchymal transition (EMT) in pancreatic cancer cells. Methods: Pancreatic cancer cell line PANC-1 cells were cultured in vitro and treated with 10 μg · L -1 transforming growth factor-β 1 (TGF-β 1), 10 μg · L -1 TGF-β 1 +5 mg · L -1 Allicin and 5 mg · L -1 Garlicin were used to treat PANC-1 cells. A blank group was also established. Morphological changes were observed under the microscope. Cell scratch assay was used to detect whether allicin can reduce the migration ability of pancreatic cancer cells. Immunofluorescence cytochemistry and Western blot were used to detect the expression of E-cadherin, Vimentin Change of expression Results: TGF-β 1 could stimulate typical morphological changes of EMT in PANC-1 cells with the up-regulation of E-cadherin expression and down-regulation of Vimentin. Scratch experiments showed that allicin can reduce the migration of pancreatic cancer PANC-1 cells. Immunofluorescence cytochemistry and Western blot results showed that compared with the blank group and the TGF-β1 group, the expression of E-cadherin in allicin group was significantly increased (P <0.05) and Vimentin was significantly decreased (P <0.05), indicating that allicin Has the effect of inhibiting PANC-1 cells to produce EMT. Conclusion Allicin can inhibit pancreatic cancer cells from EMT, which may play an important role in inhibiting tumor invasion and metastasis.
