目的探索肺血栓栓塞症(PET)大鼠肺组织不规则趋化因子(FKN)的变化及姜黄素(Cur)的干预作用。方法采用自体血栓法复制大鼠PTE模型。96只大鼠按完全随机分组方法分为6组:正常组(control组)、假手术组(sham组)、模型组(model组)、Cur低剂量组(Cur-low组)、CUR中剂量组(Cur-medium组)、Cur高剂量组(Cur-high组)。栓塞后4、72h,行HE染色观察肺病理学改变,检测肺系数、血清FKN和肺组织的FKN、肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)的mRNA水平。结果栓塞后4、72h,各组肺系数比较,差异均无统计学意义(P均>0.05)。栓塞后4h血清FKN比较,control组明显低于model组,72h后control、sham、Cur-high组均明显低于model组(P均<0.01)。Control、sham、Cur-low、Cur-medium、Cur-high组4、72h肺组织FKN、TNF-α、IL-1β的mRNA水平均明显低于model组(P均<0.01)。栓塞后72h,大鼠肺组织HE染色显示肺泡壁血管高度扩张、充血,光镜下个别大鼠查见出血性梗死灶,应用Cur后肺组织充血程度明显减轻,出现梗死的大鼠数量也有降低的趋势。结论 Cur能降低PTE大鼠肺组织的FKN水平。
Objective To explore the changes of lung tissue irregular chemotactic factor (FKN) in pulmonary thromboembolism (PET) rats and the effect of curcumin (Cur) on it. Methods Autologous thrombosis was used to duplicate rat PTE model. Ninety-six rats were randomly divided into six groups: control group, sham group, model group, Cur low group, CUR medium dose (Cur-medium group), Cur high dose group (Cur-high group). At 4 and 72 hours after embolization, the pathological changes of lung were observed by HE staining. The lung coefficient, serum FKN, FKN, TNF-α and IL-1β mRNA levels in lung tissue were detected. Results At 4 and 72 hours after embolization, there was no significant difference in lung coefficient between groups (P> 0.05). At 4h after embolization, the serum FKN level in control group was significantly lower than that in model group. After 72h, the control, sham and Cur-high groups were significantly lower than those in model group (all P <0.01). The mRNA levels of FKN, TNF-α and IL-1βin Control, Sham, Cur-low, Cur-medium and Cur-high groups were significantly lower than those in model group at 4 and 72h (P <0.01). At 72h after embolization, HE staining showed that the vascular wall of alveolar wall was highly expanded and hyperemic. Hemorrhagic infarction was detected in individual rats under light microscope. The extent of hyperemia in lung tissue was significantly reduced after administration of Cur, and the number of infarcted rats also decreased the trend of. Conclusion Cur can reduce the level of FKN in lung of PTE rats.
