目的:采用HPLC结合浊点萃取法测定大鼠血浆中的芦荟大黄素,大黄素和大黄酚。方法:使用非离子型表面活性剂Triton X-114作为萃取溶剂,甲醇和0.1%磷酸溶液为流动相进行梯度洗脱,以Kromasil C18为分析柱,柱温40℃,检测波长254nm。结果:芦荟大黄素,大黄素,大黄酚分别在0.117~15mg·L-1、0.258~16.5mg·L-1和0.126~16.2mg·L-1的范围内线性良好(r2>0.9997),3种成分的日内、日间精密度分别为2.85%、3.27%、4.84%和5.38%、6.01%、5.12%。结论:本方法简便、环保、快捷,适用于芦荟大黄素、大黄素和大黄酚在大鼠体内血药浓度的测定。
Objective: To determine aloe-emodin, emodin and chrysophanol in rat plasma by HPLC and cloud point extraction. Methods: The non-ionic surfactant Triton X-114 was used as the extraction solvent. Methanol and 0.1% phosphoric acid were used as the mobile phase for gradient elution. Kromasil C18 was used as the analytical column. The column temperature was 40 °C and the detection wavelength was 254 nm. Results: Aloe-emodin, emodin, and chrysophanol were linear in the range of 0.117-15 mg·L-1, 0.258-16.5 mg·L-1, and 0.126-16.2 mg·L-1, respectively (r2>0.9997). 3 The intra- and inter-day precision of the components was 2.85%, 3.27%, 4.84%, and 5.38%, 6.01%, and 5.12%, respectively. Conclusion: This method is simple, environment-friendly and fast. It is suitable for the determination of blood concentration of aloe-emodin, emodin and chrysophanol in rats.
