本文用提纯的小牛胸腺末端脱氧核苷酰转移酶(TdT)为免疫原,经一次融合,建立了3株能稳定分泌抗小牛胸腺TdT单克隆抗体(单抗)的杂交瘤细胞株(命名为抗TdT1、TdT2和TdT3)。3株单抗均为IgG1抗体;抗体相加试验证明抗TdT2和抗TdT3识别TdT的同一表位,而抗TdT1则识别另一表位;3株单抗识别的抗原分子量均为58.8kD;与人白血病细胞株及各型白血病细胞的反应谱3株单抗和进口抗TdT单抗基本一致。提示这3株单抗有可能替代进口抗TdT单抗,供国内使用。
In this paper, we purified three calf thymus terminal deoxynucleotidyl transferase (TdT) as the immunogen, and established three hybridoma cell lines that can stably secrete anti-calf thymus monoclonal antibody (mAb) Named anti-TdT1, TdT2 and TdT3). The three McAbs were all IgG1 antibody. Antibody addition test showed that anti-TdT2 and anti-TdT3 recognized the same epitope of TdT while anti-TdT1 recognized another epitope. The molecular weight of the three MAbs was 58.8 kD. Antibody against leukemia cell lines and leukemia cells were basically the same as that of the three McAbs and the imported anti-TdT mAb. Suggesting that these three monoclonal antibodies may replace imported anti-TdT monoclonal antibody for domestic use.
