目的研究芎芷地龙汤对偏头痛模型大鼠的镇痛作用机制。方法将48只健康雄性SD大鼠随机为生理盐水组、模型组、舒马普坦组、中药组,每组12只。皮下注射硝酸甘油制备偏头痛动物模型,经药物干预后在4h、6h取材,测定三叉神经脊束核(SPN5)和中脑导水管周围灰质(PAG)中细胞癌基因fos(c-fos)和5-羟色胺(5-HT)的阳性细胞数。结果 SPN5中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较差异有统计学意义(P<0.01);5-HT阳性细胞数增多,4h稍多于6h,与模型组比较有统计学差异(P<0.05或P<0.01)。PAG中蛋白表达:中药组c-fos阳性细胞数减少,4h多于6h,与模型组比较有统计学差异(P<0.01);中药组5-HT阳性细胞数增加更多,6h多于4h,与模型组比较有统计学差异(P<0.01)。结论芎芷地龙汤可以减少由硝酸甘油诱导的c-fos的表达,增加5-HT的表达,进一步减少SPN5神经元的兴奋性,激动5-HT下行镇痛系统,且可持续较长时间,增高痛阈,阻止痛觉信息的传递,起到镇痛作用。
Objective To study the analgesic mechanism of Xiongzhi Dilong decoction on migraine model rats. Methods Forty-eight healthy male Sprague-Dawley rats were randomly divided into normal saline group, model group, sumatriptan group and traditional Chinese medicine group, with 12 rats in each group. Subcutaneous injection of nitroglycerin in animal models of migraine, after drug intervention at 4h, 6h were drawn to determine the trigeminal nerve nucleus (SPN5) and midbrain periaqueductal gray (PAG) of the cell carcinoma gene fos (c-fos) and The number of serotonin (5-HT) positive cells. Results The protein expression of SPN5: The number of c-fos positive cells decreased in TCM group for more than 6h after 4h, which was significantly different from that in model group (P <0.01); the number of 5-HT positive cells increased more than that in 4h slightly more than 6h, Compared with the model group, there was a significant difference (P <0.05 or P <0.01). PAG protein expression: Chinese medicine group c-fos positive cells decreased 4h more than 6h, compared with the model group was statistically significant (P <0.01); Chinese medicine group 5-HT positive cells increased more, 6h more than 4h , Compared with the model group was statistically significant (P <0.01). CONCLUSION Xiongzhi Dilong decoction can reduce the expression of c-fos induced by nitroglycerin, increase the expression of 5-HT, further reduce the excitability of SPN5 neurons, stimulate the 5-HT descending analgesic system, and can last for a long time , Increase the pain threshold, prevent the transmission of pain information, play a role in analgesia.
