目的探讨胰岛素样生长因子Ⅱm RNA结合蛋白3(IMP3)、Ki-67、P16INK4 a在宫颈上皮内瘤变(CIN)及浸润性癌中的表达与临床病理意义。方法采用免疫组化S-P二步法及原位杂交方法检测20例正常宫颈上皮(NCE)、100例CIN、80例宫颈鳞状细胞癌(SCC)患者的IMP3蛋白、Ki-67及P16INK4 a蛋白的表达情况。结果IMP3蛋白在NCE、CIN(Ⅰ、Ⅱ、Ⅲ)、SCC中的阳性表达率分别为0.0%、0.0%、40.0%、71.1%、91.2%,两两比较差异均有统计学意义(P<0.05),IMP3蛋白表达与宫颈病变程度呈正相关。P16INK4 a蛋白在NCE、CIN(Ⅰ、Ⅱ、Ⅲ)和SCC中的阳性表达率分别为0.0%、52.0%、96.6%、95.5%、97.8%。SCC中IMP3及P16INK4 a的阳性表达率均与组织学分化有关(P<0.05),IMP3蛋白与P16INK4 a蛋白的表达呈正相关(r=0.390,P=0.005);Ki-67在SCC以及CIN中广泛表达,少数正常子宫颈组织中也有表达,在正常组、CIN组以及SCC组中的阳性表达率不同,其差异有统计学意义(P<0.05)。结论IMP3、Ki-67及P16INK4 a表达可为早期宫颈癌的病理诊断提供一定的实验依据,可将其作为宫颈癌转移潜能的生物学标记物之一。
Objective To investigate the expression and clinical pathological significance of insulin-like growth factor Ⅱm RNA binding protein 3 (IMP3), Ki-67 and P16INK4 a in cervical intraepithelial neoplasia (CIN) and invasive carcinoma. Methods The expression of IMP3 protein, Ki-67 and P16INK4 a protein in 20 cases of normal cervical epithelium (NCE), 100 cases of CIN and 80 cases of cervical squamous cell carcinoma (SCC) were detected by immunohistochemical SP method and in situ hybridization The expression of the situation. Results The positive rates of IMP3 protein in NCE, CIN (Ⅰ, Ⅱ, Ⅲ) and SCC were 0.0%, 0.0%, 40.0%, 71.1% and 91.2% 0.05). The expression of IMP3 protein was positively correlated with the degree of cervical lesions. The positive rates of P16INK4 a protein in NCE, CIN (Ⅰ, Ⅱ, Ⅲ) and SCC were 0.0%, 52.0%, 96.6%, 95.5% and 97.8%, respectively. The positive expression rate of IMP3 and P16INK4 a in SCC was correlated with histological differentiation (P <0.05), and the expression of IMP3 protein and P16INK4 a protein were positively correlated (r = 0.390, P = 0.005) The expression was widespread in a few normal cervical tissues. The positive expression rate was different in normal group, CIN group and SCC group, the difference was statistically significant (P <0.05). Conclusion The expressions of IMP3, Ki-67 and P16INK4a may provide some experimental evidences for the pathological diagnosis of early cervical cancer, which may be used as one of the biomarkers of cervical cancer metastasis potential.
