目的探讨黄莲提取液对人神经胶质瘤细胞(C6)体外增殖的影响及其可能的作用机制。方法用10、20、40、60μl/ml的黄连提取液分别作用C6细胞48 h后,倒置显微镜下观察C6细胞形态;作用24、48、72 h后,MTT法检测黄莲提取液对C6细胞体外增殖的影响。用40μl/ml的黄连提取液分别作用C6细胞0、20、40及60 min后,Western blot法分析C6细胞中Bcl-2和Bax蛋白的表达水平。结果倒置显微镜下观察可见,随着黄莲提取液浓度的升高,坏死细胞的数量逐渐增多;40μl/ml黄莲提取液作用48 h时的抑制率显著高于10及20μl/ml浓度组(P<0.05),与60μl/ml浓度组差异无统计学意义(P>0.05);C6细胞中Bax蛋白表达水平随黄莲提取液作用时间的延长而上升(P<0.05),Bcl-2蛋白表达水平随黄莲提取液作用时间的延长而下降(P<0.05)。结论黄莲提取液对C6细胞的增殖具有显著的抑制作用,且可能是通过Bcl-2途径实现的。
Objective To investigate the effect of extract of Radix Angelica on the proliferation of human glioma cells (C6) in vitro and its possible mechanism. Methods C6 cells were treated with Rhizoma Coptidis in 10, 20, 40, and 60 μl/ml for 48 h respectively. The morphology of C6 cells was observed under inverted microscope. After 24, 48, and 72 h, MTT assay was used to detect Coptis In vitro proliferation effects. After C6 cells were treated with 40 μl/ml Coptis extract for 0, 20, 40 and 60 min, the expression of Bcl-2 and Bax protein in C6 cells was analyzed by Western blot. RESULTS: Under the inverted microscope, the number of necrotic cells increased gradually with the increase of the concentration of the extract, and the inhibitory rate of 40 μl/ml of the extract was significantly higher than that of the 10 and 20 μl/ml groups at 48 h. P<0.05), there was no significant difference with 60μl/ml concentration group (P>0.05); the expression of Bax protein in C6 cells increased with the prolongation of the time of the effect of the extract (P<0.05). Bcl-2 protein The expression level decreased with the prolongation of the time of the effect of the extract (P<0.05). Conclusion The extract of Coptis Chinensis has a significant inhibitory effect on the proliferation of C6 cells, and may be achieved through the Bcl-2 pathway.
