应用PCR-单链构象多态分析(SSCP)银染技术分析了21例结肠癌和正常结肠粘膜组织p53基因第五~第八外显子。发现了9例(43%)的结肠癌组织出现了反映p53基因突变的异常条带。结果显示p53基因的突变与结肠癌的发生和发展有关。应用非同位素的银染方法,能简单、快速检测基因多态性,并避免同位素的危害,对结肠癌的基因诊断有潜在性应用价值
The fifth to the eighth exons of p53 gene in 21 cases of colon cancer and normal colonic mucosa were analyzed by PCR-single strand conformation polymorphism (SSCP) silver staining. In 9 cases (43%) of colon cancers, abnormal bands reflecting p53 gene mutation were found. The results showed that the mutation of p53 gene is associated with the occurrence and development of colon cancer. The application of non-isotope silver staining method can detect gene polymorphism easily and rapidly and avoid the harm of isotope, and has potential application value in gene diagnosis of colon cancer
