目的对16例来自凉山彝族自治州的HIV感染者进行亚型分析,监测该地区感染人群HIV毒株的流行情况。方法从16例HIV-1抗体阳性样本中分离外周血单核细胞(PBMC),从PBMC中提取基因组DNA,直接进行巢式PCR扩增HIVenv基因c2~v5区及gag基因p17~p24区,对PCR产物进行核苷酸序列测定和分析,确定基因亚型。结果 16例样本全部为BC重组亚型,env基因的基因距离较gag基因远,gag基因在进化上与CRF07-BC更为接近。结论凉山彝族自治州的HIV流行毒株主要为CRF07-BC。
Objective To analyze subtypes of 16 HIV-infected individuals from Liangshan Yi Autonomous Prefecture and monitor the prevalence of HIV strains in the infected population in the area. Methods Peripheral blood mononuclear cells (PBMCs) were isolated from 16 HIV-1 antibody-positive samples. Genomic DNA was extracted from PBMCs. The nested PCR was used to amplify HIV-2 v5 region and p17-p24 region of gag gene. PCR products were subjected to nucleotide sequencing and analysis to determine the genotypes. Results All of the 16 samples were BC subtype. The distance between env gene and gag gene was far, and gag gene was closer to CRF07-BC gene. Conclusion The epidemic strains of HIV in Liangshan Yi Autonomous Prefecture are mainly CRF07-BC.
