目的了解不同类型热休克蛋白(HSP)在大肠癌组织中的表达,并探讨其与大肠癌的发生、发展及生物学行为的关系。方法收集大肠癌患者标本存档蜡块48例,32例选自中国医科大学附属第二临床医院内窥镜中心,应用肠镜取材的病例标本,16例选自本院外科大肠癌手术后标本,48例标本分为两组,一组为大肠癌组织取材标本;另一组为同一大肠癌患者距癌组织10~15cm正常癌旁黏膜组织取材。应用免疫组化SABC法观察40例大肠癌组织和癌旁正常组织中HSP60、70、90α的表达,并比较期间是否存在差异性。结果大肠癌组织和癌旁正常组织粘膜中HSP60、70、90α的阳性表达率分别为54%、60%、79%和21%、17%、21%.3种HSP在癌组织与癌旁正常组织中表达差异均有显著性意义(χ2=13.653,21.333,28.800,P<0.01)。结论3种HSP可在大肠癌中呈高表达,各类型HSP的表达与大肠癌的分化程度及淋巴节转移无关,但HSP在直肠癌中的过量表达提示预后不良,可做为直肠癌预后判定监测标记。
Objective To investigate the expression of different types of heat shock protein (HSP) in colorectal cancer and to explore its relationship with the occurrence, development and biological behavior of colorectal cancer. Methods Forty-eight paraffin blocks were collected from patients with colorectal cancer, 32 cases were selected from the Endoscopy Center of the Second Affiliated Hospital of China Medical University, the cases were selected from the colonoscopy, 16 cases were selected from the surgical specimens of surgical colorectal cancer, 48 specimens were divided into two groups, one for the specimens of colorectal cancer tissue; the other for the same colorectal cancer patients from cancer tissue 10 ~ 15cm adjacent normal mucosa tissue taken. Immunohistochemical SABC method was used to observe the expression of HSP60, 70,90α in 40 cases of colorectal cancer tissues and adjacent normal tissues, and whether there was any difference between the two groups. Results The positive expression rates of HSP60, 70,90α in colorectal cancer tissues and adjacent normal mucosa were 54%, 60%, 79% and 21%, 17% and 21% There were significant differences in the expression in tissues (χ2 = 13.653, 21.333, 28.800, P <0.01). Conclusion The three HSPs are highly expressed in colorectal cancer. The expression of HSPs is not related to the differentiation and lymph node metastasis of colorectal cancer. However, the overexpression of HSP in rectal cancer suggests a poor prognosis and may be used as a prognostic marker for rectal cancer Monitoring mark.
