目的建立应用全自动生化分析仪低砷快速测定高质量浓度尿中碘含量的测定方法,实现尿碘高于300μg/L样品的直接测定,适用于高碘地区高效开展大批量尿碘的实验室检测。方法结合生化分析仪,自动进样、加试剂和比色检测的优势,进行尿中碘含量的测定,开展相应的线性范围、精密度和准确度等验证实验。结果标准曲线线性范围为300~1 200μg/L,取样量500μL,对低、中、高3种不同含量的尿样,精密度实验CV%范围分别为1.86%、1.25%、1.84%。加标回收率分别为104.0%、98.3%、104.5%(n=6),平均回收率为102.3%。结论该方法仪器操作简单,环境适应性好,检测线性范围合理;亚砷酸的使用量仅为国标法的2%,自动化实验操作减轻了检测人员的工作负担及与有毒试剂的直接接触,使得检测工作安全、高效。该方法实现了自动化检测,检测速度快,适用于高碘地区尿样碘含量的检测。
OBJECTIVE To establish a method for the rapid determination of iodine in high-concentration urine with low-arsenic by automatic biochemical analyzer, to directly measure urinary iodine above 300μg / L, suitable for high-iodine-rich laboratories Detection. Methods Combined with the advantages of biochemical analyzer, automatic sample injection, reagent addition and colorimetric detection, the determination of iodine in urine was carried out, and corresponding verification experiments such as linear range, precision and accuracy were carried out. Results The linear range of the standard curve was 300-1 200 μg / L and the sample volume was 500 μL. The CV% range of precision, low, medium and high were 1.86%, 1.25% and 1.84% respectively. The spiked recoveries were 104.0%, 98.3% and 104.5% (n = 6), respectively. The average recoveries were 102.3%. Conclusions The method is simple in operation, good in environmental adaptability and reasonable linear range of detection. The amount of arsenious acid used is only 2% of the national standard method. The automated experimental operation can reduce the workload of the inspectors and make direct contact with toxic reagents. Testing work safe and efficient. The method achieves automatic detection, fast detection and is suitable for the detection of iodine in urine samples in areas with high iodine.
