目的:研究榄香烯对人喉鳞状细胞癌细胞株Hep-2细胞荷瘤裸鼠模型移植瘤的生长抑制作用,探讨其抑瘤机制。方法:建立人喉鳞状细胞癌Hep-2细胞株裸鼠皮下移植模型。应用榄香烯治疗,观察移植肿瘤和裸鼠生长情况,计算肿瘤抑制率;逆转录PCR检测肿瘤组织中血管内皮生长因子C(VEGF-C)和血管内皮生长因子受体3(VEGFR-3)mRNA表达情况。结果:榄香烯对人喉鳞状细胞癌细胞株Hep-2细胞荷瘤裸鼠模型移植瘤的增殖具有明显抑制作用,抑瘤率为52.24%。在实验组和对照组之间,鼠净重、瘤重和瘤体积相比,差异有统计学意义(均P<0.01);实验组VEGF-C和VEGFR-3mRNA的表达低于对照组(P<0.01)。结论:榄香烯可显著抑制人喉鳞状细胞癌裸鼠模型肿瘤的生长,其机制与榄香烯抑制VEGF-C和VEGFR-3的表达有关。
OBJECTIVE: To study the inhibitory effect of elemene on the growth of transplanted human laryngeal squamous cell carcinoma cell line Hep-2 in nude mice and to explore its antitumor mechanism. Methods: Human laryngeal squamous cell carcinoma Hep-2 cell line was subcutaneously transplanted into nude mice. The growth of the transplanted tumor and the nude mice were observed and the tumor inhibition rate was calculated by using elemene treatment. The expressions of VEGF-C and VEGFR-3 were detected by reverse transcription polymerase chain reaction (RT-PCR) mRNA expression. Results: Elemene had a significant inhibitory effect on the proliferation of transplanted human laryngeal squamous cell carcinoma cell line Hep-2 in nude mice model with tumor inhibition rate of 52.24%. There were significant differences in the net weight, tumor weight and tumor volume between the experimental group and the control group (all P <0.01). The expression of VEGF-C and VEGFR-3 mRNA in the experimental group was lower than that in the control group (P < 0.01). Conclusion: Elemene can significantly inhibit the growth of human laryngeal squamous cell carcinoma in nude mice. The mechanism is related to the inhibition of the expression of VEGF-C and VEGFR-3 by elemene.
